Non-Uniform Sampling (NUS) for Everyday Use: Sharper Spectra in Less Time

چکیده
This application note demonstrates non-uniform sampling (NUS) acquisition and processing for routine measurement of 2D NMR spectra using tools in Agilent VnmrJ 4 software. It shows that NUS techniques can be applied easily and reliably to small molecule NMR measurements, yielding 2D spectra with comparable digital resolution to those obtained by traditional linear sampling methods, typically with a twoto four-fold reduction in experiment time. It also shows that NUS can enable higher resolution spectra to be obtained in a given measurement time compared to traditional linear sampling and Fourier Transform (FT) methods. Joint non-uniform sampling of all incremented time delays for quicker acquisition in protein relaxation studies
NMR relaxometry plays crucial role in studies of protein dynamics. The measurement of longitudinal and transverse relaxation rates of [Formula: see text]N is the main source of information on backbone motions. However, even the most basic approach exploiting a series of [Formula: see text]N HSQC spectra can require several hours of measurement time. Standard non-uniform sampling (NUS), i.e. ran...
متن کاملEXtended ACquisition Time (EXACT) NMR-A Case for 'Burst' Non-Uniform Sampling.
A strong case exists for the introduction of burst non-uniform sampling (NUS) in the direct dimension of NMR spectroscopy experiments. The resulting gaps in the NMR free induction decay can reduce the power demands of long experiments (by switching off broadband decoupling for example) and/or be used to introduce additional pulses (to refocus homonuclear coupling, for example). The final EXtend...
متن کاملDeterministic schedules for robust and reproducible non-uniform sampling in multidimensional NMR.
We show that a simple, general, and easily reproducible method for generating non-uniform sampling (NUS) schedules preserves the benefits of random sampling, including inherently reduced sampling artifacts, while removing the pitfalls associated with choosing an arbitrary seed. Sampling schedules are generated from a discrete cumulative distribution function (CDF) that closely fits the continuo...

Two-dimensional covariance (COV2D) spectroscopy offers an alternative approach to 2D Fourier transformation (FT2D) to obtain homo-nuclear correlation NMR spectra. Considerable saving in experimental time, without loss of resolution, can be achieved by incorporating COV2D with: (i) uniform sampling with a t1 cut-off (CUO) or non-uniform sampling (NUS), and (ii) constant or Gaussian accumulation ...
متن کاملArtifacts in time-resolved NUS: A case study of NOE build-up curves from 2D NOESY.
Multidimensional NMR spectroscopy requires time-consuming sampling of indirect dimensions and so is usually used to study stable samples. However, dynamically changing compounds or their mixtures commonly occur in problems of natural science. Monitoring them requires the use multidimensional NMR in a time-resolved manner - in other words, a series of quick spectra must be acquired at different ...
